چکیده

Bernheim (2), studying the properties of the “citric dehydrogenase” system, noted that aconitic acid (presumably the trans isomer) inhibited the reduction of methylene blue by citric acid. More recently, Morrison and Still (3), in a short report, claimed that truns-aconitic acid inhibited competitively the reaction cis-aconitate c citrate, catalyzed by the aconitase of rhubarb leaf. The present studies show that trans-aconitate is a competitive inhibitor of aconitase of animal tissues and that trans-aconitate inhibits the respiration of tissue slices and causes the accumulation of citrate by surviving kidney cortex and liver slices.

Aconitase and citric acid fermentation by Aspergillus niger.

In view of the often-cited theory that citric acid accumulation is caused by an inhibition of aconitase activity, the equilibrium of the reaction of aconitase was investigated by comparing in vivo steady-state concentrations of citrate and isocitrate in Aspergillus niger grown under various citric acid-producing conditions. With the equilibrium catalyzed by the A. niger enzyme in vitro, similar...

The Mechanism of Aconitase Action EVIDENCE FOR AK EKZYR’IE ISOMERIZATIOK BY STUDIES OF IKHIBITIOK BY TRICARBOXYLIC ACIDS *

Tricarboxylic acids were shown to be both competitive and noncompetitive inhibitors of aconitase with the inhibition pattern dependent upon which substrate was used. Tricarballyate and fluorocitrate were linear competitive inhibitors when citrate or isocitrate was the substrate but noncompetitive inhibitors when cis-aconitate was the substrate. transAconitate was a linear competitive inhibitor ...

The inhibition of plant aconitate hydratase (aconitase) by fluorocitrate.

Fluoroacetate is a potent poison of animals. It is metabolized to fluorocitrate, which is a powerful inhibitor of mammalian aconitate hydratase [aconitase; citrate (isocitrate) hydrolyase] (Peters, 1952). Fluoroacetate and fluoroacetamide, as systemic insecticides for plants, are taken up by the roots or absorbed by the leaves and are then translocated to other parts of the plant. The plants sh...

Distinct reactions catalyzed by bacterial and yeast trans-aconitate methyltransferases.

The trans-aconitate methyltransferase from the bacterium Escherichia coli catalyzes the monomethyl esterification of trans-aconitate and related compounds. Using two-dimensional (1)H/(13)C nuclear magnetic resonance spectroscopy, we show that the methylation is specific to one of the three carboxyl groups and further demonstrate that the product is the 6-methyl ester of trans-aconitate (E-3-car...

3-Isopropylmalate is the major endogenous substrate of the Saccharomyces cerevisiae trans-aconitate methyltransferase.

The Saccharomyces cerevisiae Tmt1 gene product is the yeast homologue of the Escherichia coli enzyme that catalyzes the methyl esterification of trans-aconitate, a thermodynamically favored isomer of cis-aconitate and an inhibitor of the citric acid cycle. It has been proposed that methylation may attenuate trans-aconitate inhibition of aconitase and other enzymes of the cycle. Although trans-a...
